Would it be possible for me to arrange a trip to Robben Island on Day 3 instead of the additional optional excursion to the winelands, since this seems to be covered on day 5 & 6?

You will be free to organise anything you want on day 3 if you do not wish to go on the excursion as the excursion to the Winelands is only optional.
